OPTIMIZATION PROCEDURE OF METAL-AFFINITY
CHROMATOGRAPHY ON SORBENTS, CONTAINING IONS OF Ti (IV) FOR ISOLATION OF PHOSPHONYLATED PEPTIDES
|
The present article describes the optimization procedure of metal-affinity chromatography on sorbents, containing ions of Ti (IV) (Supel-Tips Ti) for separation of peptides from the digest of human serum albumin,
incubated with sarin. It is shown that changing the conditions recommended by the manufacturer, namely sorption of alkaline conditions rather instead of acidic, and elution of 0.1 % TFA in 60 % acetonitrile instead of 0.5 % pipe-ridine
results in a more efficient isolation of phosphonylation peptides from the samples |

INTERPRETATION OF QUANTITATIVE REAL-TIME PCR ANALYSIS RESULTS BASED ON THE PROBABILISTIC MODELS |
The present work develops our results obtained in recent studies [1–3] employing stochastic imitation modeling to analyze real-time PCR kinetics and DNA
quantitative assay performance. In particular, PCR threshold cycle probability distribution depending on initial DNA copy number was estimated, reaction kinetics was studied, quantitative assay errors were
analyzed. In the present paper, a number of topics related to the quantitative assay results interpretation is discussed: estimation of copy number (n) in the sample based on a series of quantitative assay results (ki, i = 1, 2,…, m)
for a number of assay runs; obtaining estimates ki; interpretation of negative or false-negative assay results in the case of multiple (m) runs with no copies found. |

COMPOSITION OF THE CALIBRATION LINE
DURING DNA FRAGMENT ANALYSIS
|
It’s proposed to use an approximation of the tabulated dependence of the peaks position of the internal standard on the time by a polynomial of the fourth degree as an analytic function. This analytic function permits to plot a calibration line to determine the optimal length of the analyzed DNA fragments within the calibration range and beyond its range. |

OPTIMUM DISCRETIZATION OF VELOCITY SCALE IN MÖSSBAUER SPECTRUM MEASUREMENTS
|
The paper describes
two approaches for selecting of the optimum channel widths
by a discretization of the velocity scale in the process of
Mössbauer spectra accumulation by means of multichannel
accumulating mechanisms. The first approach is based on an
analysis of the Kotel’nikov—Shannon’s frequency criterion,
the second approach is based on an analysis of errors that
arise in the case of a differential representation of the
spectrum. In this case, the analysis is conducted in two stages:
in the case of the first step the shape distortion of the
ideal spectrum due to discretization of the velocity scale
is considered, but in the case of the second step the influence
of the statistical scatter in the spectrum by introducing
of the "spectrum quality" parameter (signal/noise relation)
are taken into account. The mathematical expressions are derived
for estimation the width of the channel for the each approach. |

THE METHOD OF CALCULATION OF POTENTIALS FOR THE FIELD DETERMINATION ELEMENTS OF NON-LINEAR ION MIRROR |
À method for calculating of the non-uniform electrostatic field of the mass reflection reflector is described. The method allows to calculate the axial potential distribution in the reflector theoretically eliminating dispersion of the total time of flight for axial achromatic ion packets in a given range of the initial energy of the ions. By the method of the numerical simulation the analysis of the errors influence for the field physical synthesis close to the real reflector on the focusing accuracy is carried out. The calculation procedure of the reflector with an inhomogeneous field is proposed. The description of the potentials optimization algorithm for field-determination elements is given. The recommendations to reduce execution time of the algorithm are given. The design used for verification of the optimization algorithm is described; the results of simulation of the focusing procedures for ions of different masses and different initial energy spreads are given. |

A STUDY ON AGGREGATES CLUSTER STRUCTURE OF POLYMERIC
MICROGLOBULES ON THE CUTTING SURFACE OF MONOLITHIC FUSED-SILICA COLUMNS FOR CAPILLARY
ELECTROCHROMATOGRAPHY
|
On the basis of a cluster lattice model, the cross-sections of monolithic sorbents in fused-silica microcolumns for capillary electrochromatography were studied. The columns were obtained by polymerization of methacrylate monomers in solution under the influence of accelerated electrons. It was shown that the size distribution and packing density of microglobules on the surface cross-sections depend on the composition of a mixture of monomers and the content of porogenic solvents. At the threshold degree of surface coverage by globules, there is a fractal cluster function, for which the radial density distribution g (R) decreases with increasing radius R as a power law. An infinite cluster of pores, with a finite probability of the injected fluid flowing through the inter-connected free knots or percolated pores of the monolithic sorbent, is formed near the threshing surface coverage of a cross-section by microglobules. |

IMPACT OF STRUCTURIZATION IN COLLOIDAL SOLUTION
OF FERROMAGNETIC NANOPARTICLES ON DEMAGNETIZATION FACTOR OF THE SAMPLE
|
In experiment, the effective coefficient of demagnetization the sample of colloidal solution ferromagnetic nanoparticles was measured by method of NMR.
This solution was put in two long cylindrical tubes parallel to intensity of external magnetic field and having diameters are 10 times less than the height of the liquid. It is found that the measured coefficient of
demagnetization N ≈ 0.09, while in theory N = 0.02. The effect is explained by the presence of conglomerates of nanoparticles. |

THE INFLUENCE RESEARCH OF TEMPERATURE DEFORMATION
ON PRESSURE COEFFICIENT OF RESISTANCE OF THE SmS
THIN POLYCRISTALLINE FILMS
|
The article presents the results of the researches, dedicated to the temperature dependence of the pressure electrical resistivity coefficient (PRC) of the baroresistor on the ground of thin film
of the samarium monosulphide (SmS) on the glass substrate (SiO2) within the range of 77–350 K. The feature as a sharp maximum is observed throughout the temperature dependence of the PRC module.
This maximum can be explained by the growing activity of electrons conductivity from the donor level, which energy depends on the temperature deformation, appearing as the result of the temperature coefficients difference between linear
dilatation of the film and glass substrate. The calculation of the activation energy according to this donor level is carried out. On the basic of the work results the acceptable for the technical using the low temperature border for tenso- and
baroresistor operation on the ground of the SmS films equal to the ~150 Ê is determined.
