BIOSYNTHESIS OF TRANSMEMBRANE PHOTO-TRANSFORMING PROTEIN [2Í]BACTERIORHODOPSIN, LABELLED WITH DEUTERIUM ON RESIDUES OF AROMATIC AMINO ACIDS [2,3,4,5,6-2H5]Phe, [3,5-2H2]Tyr, [2,4,5,6,7-2H5]Trp

The microbial micropreparative synthesis of transmembrane photo-transforming protein bacteriorhodopsin (output 8-10 mg), labeled with deuterium on residues of [2,3,4,5,6-2H5]phenylalanine, [3,5-2H2]tyrosine, and[2,4,5,6,7-2H5]tryptophan was carried out that is important for nanoindustry of new modern domestic photo-transforming nanomaterials and molecular bioelectronics. The protein was extracted from membranes of photoorganoheterotrophic halobacterium Halobacterium halobium by cellular lysis in distilled water, processing of bacterial biomass by ultrasound, alcohol extraction of low and high-weight molecular impurities, cellular RNA, carotenoids and lipids, with the subsequent solubilization of final product with 0.5 % SDS-Na and fractionation by methanol. The homogenity of the synthesized product, and the selectivity of deuterium encorporation into the molecule was proved by combination of preparative and analytical protein methods including elecrtophoresis in 12.5 % PAAG with 0.1 % SDS-Na, gel filtration chromatography on Sephadex G-200, reverse-phase HCLP, electron impact mass-spectrometry of methyl esters of N-Dns-derivatives of amino acids.

STUDY OF THE SPECKLE STRUCTURE INFLUENCE ON THE FORMATION OF THE INTERFERENCE SIGNAL AND MEASUREMENT ERROR

Study of the influence of the speckle structure at the reflection from a diffuse surface on the formation of the interference signal envelope is performed, and measurement error is estimated. Properties of speckle fields, which are the key in the study of the characteristics of coherently bounded systems to control surfaces, are examined. Sources of measurement error are identified. Methods of processing the interference signal are considered, and the model of a priori evaluation of measurement error is suggested.

FEASIBILITY AND PRACTICAL SUPPLEMENT OF SOLUTION METHODS FOR MATHEMATICALLY INCORRECT INVERSE PROBLEM OF ELLIPSOMETRY.
2. ON SOLUTION OF INVERSE PROBLEM ON MULTIPLE SET OF LIGHT BEAM ANGLE OF INCIDENCE

The method for the mathematically incorrect inverse task of ellipsometry, based on the use of some set of sets angles of incidence of a light bunch on the sample is considered is discussed. It was shown, that the solution of the inverse task on set of angles of incidence results in a relative simplicity and reliability of the process of solution, and also to increased accuracy with which all four parameters of reflecting system with a super thin film are defined. The offered method of the solution allows to overcome the essential difficulties arising, because of a midget thickness of a film, and in an ideal case when there are no experimental errors, but the traditional approach to the solution of an inverse problem concerning all 4 parameters of single-layered reflecting system is used.

DEFORMATIONS AS A TOOL FOR DEFECT LEVELS CONTROL.
1. Zero-charged Te-antisite in CdTe

The properties of zero-charged Te-antisite (Te0Cd), have been examined using Ab-initio calculations. Defect energy levels and its occupancies were determined and symmetry reduction due to Jahn—Teller distortion seen. The Jahn—Teller distorted configuration places the 2A1a defect level closer to the valence band. This defect level position coincides with "unknown deep donor" position measured in some experiments. Partial densities of states and band structures have been calculated and analyzed. Also, the influence of three types of deformations (1D, 2D and 3D) on the defect energy levels and formation energies was investigated. It was found that the 2D deformation is the most effective for opening up the band gap of the semiconductor as well as for energetically separating the 2A1a and Ea defect levels.

ABOUT SOME ESTIMATIONS FOR CRITERION OF ONSET OF MOLECULAR IONS FRAGMENTATION IN ELECTROGASDYNAMIC FIELDS

Analytical estimations are obtained for rate constant of molecular ions dissociation at their motion in a gas medium under strong electric field. Deviation of ions velocity distribution from Maxwell thermal distribution is considered. This can be made in terms of effective ion temperature depending on electric field strength. Theoretical estimates obtained enable to evaluate the number of ion fragmentation collisions in the drift length and make it possible to estimate the field strength of fragmentation onset.

LANDAU RELATIONSHIP FOR THE TWO-SIDE PROFILE OF THE ONE-DIMENSIONAL POTENTIAL DISTRIBUTION OF THE MECHANICAL OSCILLATOR WITH THE PREDEFINED DEPENDENCE OF THE PERIOD OF OSCILLATIONS FROM ENERGY

The Landau relationship is considered which enables to reconstruct the symmetrical potential well for the one-dimensional mechanical oscillator using the predefined dependence of its period from energy as the input data. It is shown how this expression can be modified to produce non-symmetrical smooth potential wells for the same purpose. It is confirmed that there are no one-dimensional potential distributions with flat bottoms and/or with non-monotonic behavior which produce the ideally isochronous oscillators (i.e., the oscillators where the period of oscillations is the same for any energy at least for some energy ranges). The application of these results to time-of-flight mass spectrometers and FTR-MS electrostatic traps is discussed.

ON THE AVERAGE FORCE INFLUENCING THE INCLUSIONS IN SYSTEMS WITH THE DISTRIBUTED AND THE CONCENTRATED PARAMETERS

Similarity of concepts of the average force, accepted in electrodynamics (pressure of light) and in acoustics (radiating pressure), and also forces of vibrations (string) and average force of the influence on systems with the concentrated parameters, moving in quickly changing fields is stressed. On movement of particles in quickly changing fields, as an example, the identity of definition of average forces in such system and radiating pressure in acoustics is shown. Besides, on a simple example possibility of transportation of particles in points of minimum of potential in an acoustic field by analogy with previously described similar possibility is shown in systems with the concentrated parameters.

MULTIWAVE ILLUMINATOR FOR PHOTOMETRIC STUDIES OF SINGLE CELLS

Technical documentation was elaborated and a prototype of a multiwave illuminator was built, the use of which provides higher sensitivity and precision in microphotometric studies of single cells, in comparison with modern industrial analogs. The improvement of the designed illuminator characteristics was obtained as a result of the use of emitting semiconductor diodes as light sources, which ensured significant decrease of the background emission level and the deviation of the excitation light intensity compared to lamp sources. The control of the illuminator emitting wavelength is analogous, and the diode ignition control is carried out via a TTL-port. A computer control program via a USB-port enables the user to provide needed values of emitting diode cur-rents and required parameters of the illuminator ports, together with storing all the ratings in a file.

HIGH-POWER LASER DIODES EMITTING AT 808 nm. 3. WAYS TO INCREASE THE OPTICAL OUTPUT POWER

This work continues the reviews on the mechanisms limiting the output optical power of semiconductor laser diodes reported in [1, 2]. Here we consider possible ways for improving the optical power. Among them, the most effective ways are the use of large-optical cavity heterostructures, advanced coating of the laser resonator mirrors, and improving the heat sink of the laser diodes. Specific approaches suggested in each of the above directions are discussed in the paper. Assessment of the effectiveness and/or potential of the approaches is given as well.
