METHOD ECD / ETD FRAGMENTATION OF THE PHOSPHOPEPTIDES IN MODERN TANDEM MASS SPECTROMETRY

The paper presents features and the application of peptides fragmentation method in tandem mass spectrometry, in particular fragmentation techniques CID, ECD and ETD, and their combinations. Mechanism of fragmentation, sample preparation methods and sample introduction in relation to the method of ECD / ETD are analyzed. The possibility to use common proteomic databases for solving phosphoproteomic tasks is shown. The conclusions about the effectiveness of CID / ETD fragmentation methods combination to expand amino acid sequence restore capacity of the phosphopeptides are made.

ANALYSIS OF CYANOTOXINES USING AN LTQ ORBITRAP (THERMO FINNIGAN): THE OPTIMIZATION OF MASS-SPECTROMETRY METHOD

Our current research includes the optimization of methods for detection, identification and occurring ñyanobacterial toxines ("cyanotoxines"), which are the most ubiquitously found hazardous substances in surface waters. The detection limits of the optimized method for cyanotoxines determination are lower than a provisional Guideline Value for cyanotoxines concentration in drinking water provided by the World Health Organisation (1 μg/L).

APPLICATION OF CHROMATO-MASS-SPECTROMETER OF HIGH RESOLUTION LTQ ORBITRAP FOR DETERMINATION OF PERFLUORINATED ACIDS IN NATURAL WATER WITH THE TRADITIONAL SOLID-PHASE AND METALL-AFFINE SORBENTS: METHOD DEVELOPMENT AND OPTIMIZATION

Our current research includes the description of optimization procedure of the mass-spectrometry method for detection of perfluoroalkyl acids in surface waters. The principle possibility of extracting perfluorooctansulfonate from water matrix by metal-affinity chromatography on the new sorbents containing iron (III) ions was shown. The optimized method of chromato-mass-spectrometry analysis provides low limits of detection below 1ng/L that allows the accurate quantification of perfluoroalkyl acids in water samples.

IDENTIFICATION OF CYANIDE ADDUCT WITH HUMAN SERUM AL-BUMIN USING MASS-SPECTROMETRY

MALDI mass spectrometry was used to identify cyanide adducts with human serum albumin. Peptide with MH+ 999.44 Da was detected in the tryptic digestion of human serum albumin, incubated with cyanide (sodium and potassium salts). As a result SCN adduct at Cys-34 are formed. The sequence Cys-Pro-Phe-Glu-Asp-His-Val-Lys was confirmed by HPLC-MS-MS analysis on ion trap.

THE DEVELOPMENT OF HPLC-ESI-MS(+) METHOD FOR URSODEOXYCHOLIC ACID ANALYSIS

An analytical method for ursodeoxycholic acid has been developed based on high performance liquid chromatography separation coupled to an electrospray (+) time-of-flight mass-spectrometer. Data about the change of UDCA concentration after intragastric injection in rat serum depending on time are obtained.

ION’S SEPARATION IN THE STATIC FIELDS: QUADRUPOLE ELECTRIC AND HOMOGENEOUS MAGNETIC

The behaviour of ion packet in the crossing static electromagnetic fields is investigated in the article. Homogenous transverse magnetic field is applied to quadrupole analyser which works in the static mode. A number of finite trajectories and the conditions for keeping ions in the canal of the device are determined by analytical methods. The initial conditions and parameters of the fields are formulated when the system works as an ion’s trap or farvitron.

NEW DYNAMIC MASS SPECTROMETER WITH ELECTRIC IMPULSE

Charge particle optics scheme of dynamic mass spectrometer is suggested. It is based on new ideas of mass analysis. Ions of one energy are subjected to a number of short electric impulses in Δ-transformer, and as a result ions are distributed on energies. Energy spectrum recorded by energy analyzer "Modification of arch" is strictly connected with mass spectrum. Different variants of charge particle optics track are offered. Some estimations are done and perspective of dynamic mass spectrometry of new type is discussed.

STUDY OF THE SPECIFIC PROPERTIES OF MULTIMOLECULAR REGULAR SORBENT Fe(III)

Now the problem of specific separation of a variety of substances from complex mixtures, including biological samples is very acute. One of the methods that allows to solve such problems is the metal-affinity chromatography. Historically, it was used for the enrichment of phosphorus-containing peptides, but it seems possible to use it for the enrichment of other classes of substances. In the paper the specific properties of a new regular multimolecular sorbent based stearate iron (III) were discussed. It was shown that by using metal-affinity chromatography on RMMS Fe(III) such types of compounds as organophosphorus (phosphorylated peptides, phosphonylated peptides, pesticides), fluorine acid (perfluorooctane sulfonic acid), cyanobacteria toxins (microcystin-RR ) can be successfully identified and analyzed.

SYNTHESIS AND CHARACTERIZATION OF NANOREGULAR SORBENTS BASED ON ZIRCONIUM OXIDE

Application of a new method for the synthesis of ultrafine zirconium oxide from the aqueous solution with microwave heat treatment yielded particles with dimensions less than at 10 nm. High specific surface area of the produced zirconia provided a good selective sorption both of model objects (colorant Congo red) and real biological substances (casein).

STUDY OF ELECTROKINETIC PROPERTIES OF REGULAR MULTIMOLECULAR SORBENT BASED ON TRIVALENT METALS STEARATES

By the methods of streaming potential and microelectrophoresis electrokinetic properties of regular multimolecular sorbents based on metallic stearates were investigated. It was shown that for two different types of objects such as Langmuir—Blodgett films transferred onto a solid substrate and collapsed monolayers, dependences of the electrokinetic potential on pH of the electrolyte solution are practically identical, which makes the collapsed monolayers of regular multimolecular structures similar to Langmuir—Blodgett films. The values of the isoelectric point for a number of trivalent metal stearates were determined. Basing on these dependences conditions of metal-affinity chromatography sorbents on regular multimolecular were proposed.

APPLICATION OF HIGH-RESOLUTION MELTING ANALYSIS AND MALDI MINISEQUENCING FOR POLYMORPHISM GENOTYPING OF HUMAN PARAOXONASE-1 GENE

High-resolution melting analysis (HRMA) and MALDI minisequencing were used to perform single nucleotide polymorphism (rs854560 and rs662) genotyping of human paraoxonase-1 gene. The allelic frequencies (66.0 % for L-allele and 71.8 % for Q-allele) obtained from genotyping of 94 habitants of Kirov region were similar to allelic frequencies of Europeans and Caucasian Americans. The experimental data confirmed the applicability of HRM analysis and MALDI minisequencing for geno-typing of human paraoxonase-1 L55M and Q192R single nucleotide polymorphisms.

TEST OF STRAIN GAGES BASED ON SmS SEMICONDUCTOR THIN FILMS

The authors propose a method for determining the gage factor of strain gages based on polycrystalline films of samarium monosulphide (SmS) without loss of performance properties. We derive the relation between gage factor K and pressure coefficient of resistance under hydrostatic compression B (barorezistor mode). It is show that the correlation between K and B is determined only by elastic constants of strain sensitive layer of polycrystalline SmS and elastic constants of substrate on which it was formed.
